Effect of Bambusae Caulis in Liquamen on the Synthesis of Type ¥³ collagen and Laminin during Proliferation and Differentiation of 3T3-L1 Cells


Abstract
Objectives : The purpose of this research was to investigate effects of Bambusae Caulis in Liquamen (BCL) on the synthesis of basement membrane proteins during proliferation and differentiation of 3T3-Ll cells. We have analyzed synthetic amounts of basement membrane components such as type IV collagen and laminin.
Methods : 3T3-Ll cells grown on 24-well plates were cultured for 24h in the presence or absence of BCL. These cultures were metabolically labeled by incubating at 37 C for 4h in methionine free MEM containing [i5Slmethionine after incubation in the same medium without [:"S]methionine.
Results : BCL stimulated the synthesis and secretion of type IV collagen from both 3T3-Ll preadipocytes and adipocytes. The synthesis and secretion of laminin was not affected by BCL. The continuous addition of BCL markedly stimulated cell growth and increased cell density.
Conclusions : BCL stimulated the synthesis and secretion of type IV collagen from both 3T3-Ll preadipocytes and adipocytes. These results suggest an important role for type IV collagen in adipocyte hfferentiation.
